چکیده
Murine C3H10T1/2 cells were used as a model of mesenchymal stem cells (MSCs) in this study to investigate their capability of proliferation and differentiation in the miniaturized core of small (~100 μm) alginate-chitosan-alginate (ACA) microcapsules. The results show that the cells can survive and proliferate well in the microcapsules, particularly those with a liquid core. Moreover, successful adipogenic, osteogenic, and chondrogenic differentiation of the encapsulated C3H10T1/2 cells were confirmed. Collectively, the small ACA microcapsules are a promising carrier for MSC-based medicine.

A Novel Core-Shell Microcapsule for Encapsulation and 3D Culture of Embryonic Stem Cells.
In this study, we report the preparation of a novel microcapsule of ~ 100 μm with a liquid (as compared to solid-like alginate hydrogel) core and an alginate-chitosan-alginate (ACA) shell for encapsulation and culture of embryonic stem (ES) cells in the miniaturized 3D space of the liquid core. Murine R1 ES cells cultured in the microcapsules were found to survive (> 90%) well and proliferate t...
متن کاملEncapsulation of factor IX–engineered mesenchymal stem cells in fibrinogen–alginate microcapsules enhances their viability and transgene secretion
Cell microencapsulation holds significant promise as a strategy for cellular therapies; however, inadequate survival and functionality of the enclosed cells limit its application in hemophilia treatment. Here, we evaluated the use of alginate-based microcapsules to enhance the viability and transgene secretion of human cord blood-derived mesenchymal stem cells in three-dimensional cultures. Giv...
متن کاملImproving the Growth Rate of Human Adipose-Derived Mesenchymal Stem Cells in Alginate/Gelatin Versus Alginate Hydrogels
Background: Expansion and differentiation of stem cells relies on the soluble materials as well as the physical conditions of their microenvironment. Several methods have been studied in attempt to enhance the growth and differentiation rates of different adult stem cells extracted from different sources. Objectives: The purpose was to improve the three-dimensional (3D) culture condition of th...
متن کاملInjectable alginate-microencapsulated canine adipose tissue-derived mesenchymal stem cells for enhanced viable cell retention
The purpose of this study was to establish an optimized protocol for the production of alginate-encapsulated canine adipose-derived mesenchymal stem cells (cASCs) and evaluate their suitability for clinical use, including viability, proliferation and in vivo cell retention. Alginate microbeads were formed by vibrational technology and the production of injectable microbeads was performed using ...
متن کاملCulture of mesenchymal stem cells derived from equine synovial membrane in alginate hydrogel microcapsules
Background: Mesenchymal stem cells derived from the synovial membrane (MSCSM) have a greater potential for joint regeneration, besides the capacity for chondrogenic differentiation, since they are a source closer to the chondrocytes. This study aimed to cultivate and evaluate viability and differentiation of MSCSM encapsulated in a three-dimensional alginate hydrogel (HA) scaffold. Samples of t...
